利用大鼠胚胎中脑神经细胞进行原代培养(微团试验)研究了氟化钠(NaF)的细胞毒性及其对神经元分化的影响。细胞毒性试验提示NaF的半数存活抑制浓度(ICV50)为15.0μg/ml。染毒组的细胞集落形成率降低,细胞体积小,细胞间神经细胞纤维减少,其中前者呈剂量-效应关系,表明NaF可抑制胚胎神经细胞的分化,半数分化抑制浓度(ICd50)为3.0μg/ml,半数存活抑制浓度与半数分化抑制浓度的比值(v/d)大于5.0。本研究表明抑制细胞分化可能是NaF致发育危害的重要作用机理
The primary cultures of rat embryo midbrain neurons (micellar assays) were used to investigate the cytotoxicity of sodium fluoride (NaF) and its effect on neuronal differentiation. Cytotoxicity assays suggest that the half-life inhibitory concentration (ICV50) of NaF is 15.0 μg / ml. The rate of colony formation was decreased, the cell volume was small and the number of intercellular nerve fibers was decreased. The former showed a dose-effect relationship, indicating that NaF inhibited the differentiation of embryonic nerve cells. The IC50 was 3.0μg / Ml, the ratio of the half survival inhibition concentration to the half inhibition concentration (v / d) is greater than 5.0. This study shows that inhibition of cell differentiation may be an important mechanism of action of NaF-induced development
