目的利用Meta分析探讨ADAM33基因T1、V4多态性位点与哮喘易感性关系。方法检索EMBASE,Medline,springerlink,CNKI,万方,收集研究ADAM33基因T1、V4多态性位点与哮喘易感性的文献进行Meta分析。利用Revman5.0软件和STATA10.0软件进行统计学分析。结果根据纳入排除标准,总计纳入14篇文献进行研究,累计病例和对照数各自分别为2 479、3 577例和2 760、4 205例。哮喘组和对照组ADAM33基因T1、V4多态性位点的突变基因型频率同野生型纯合子基因型频率的比值比(OR)的合并OR值分别为1.10(95%CI:0.86~1.41)和1.38(95%CI:1.12~1.70)。亚组分析认为,ADAM33基因的T1多态性位点的变异对我国人群具有增加患哮喘的风险,ADAM33基因的V4多态性位点的变异对我国及其他亚洲人群具有增加患哮喘的风险,但对于欧美洲人群而言,均不具有增加患哮喘的风险。Egger线性回归分析提示ADAM33基因型相关文献没有发表偏倚。结论 ADAM33基因V4多态性位点具有增加患哮喘的风险,但T1多态性位点与哮喘易感性无统计学意义。
Objective To explore the relationship between polymorphism sites of ADAM33 gene T1 and V4 and the susceptibility to asthma by Meta-analysis. METHODS: EMBASE, Medline, springerlink, CNKI and Wanfang were collected for Meta-analysis of the literature on the study of ADAM33 T1 and V4 polymorphisms and asthma susceptibility. Using Revman5.0 software and STATA10.0 software for statistical analysis. Results According to inclusion criteria, a total of 14 articles were included in the study. The cumulative cases and controls were respectively 2 479,3 577 cases and 2 760 445 cases. The pooled odds ratio (OR) of the odds ratio (OR) between the genotype frequencies of T1 and V4 polymorphisms of ADAM33 gene and wild type homozygote in asthma group and control group were 1.10 (95% CI: 0.86-1.41) And 1.38 (95% CI: 1.12-1.70). In the subgroup analysis, the variation of T1 polymorphism site of ADAM33 gene may increase the risk of asthma in Chinese population. The mutation of V4 polymorphism site of ADAM33 gene may increase the risk of asthma in China and other Asian populations, However, for the European population, neither increase the risk of asthma. Egger linear regression analysis revealed no publication bias in ADAM33 genotype-related literature. Conclusion ADAM33 gene V4 polymorphism increases the risk of asthma, but there is no statistical significance in the susceptibility of T1 polymorphism to asthma.
