目的检测Heat shock protein 70-2(Hsp70-2)及14-3-3zeta在膀胱尿路上皮癌(bladder urothelial carcinoma,BUC)组织中的表达,探讨两者在肿瘤发展中的作用及临床意义。方法收集人膀胱尿路上皮癌标本48例,膀胱尿路上皮癌旁组织标本20例,利用免疫组织化学S-P法、Western blot和逆转录聚合酶链反应(RT-PCR)检测Hsp70-2、14-3-3zeta在蛋白水平和mRNA水平的表达,分析两者与膀胱尿路上皮癌的病理分级、临床分期等因素间的关系。结果 3种检测方法均显示Hsp70-2、14-3-3zeta在癌旁组织中低表达,在膀胱尿路上皮癌组织中高表达,其表达量随病理分级程度及浸润深度增高而增高,组间比较差异均有统计学意义(P<0.05)。Hsp70-2与14-3-3zeta蛋白在膀胱尿路上皮癌中的表达均增高,两者呈正相关(r=0.516,P<0.05)。结论 Hsp70-2与14-3-3zeta在膀胱尿路上皮癌组织中高表达,与病理分级,临床分期呈正相关,且两者之间也呈正相关,提示两者在肿瘤发生、发展过程中起重要作用。
Objective To detect the expression of heat shock protein 70-2 (Hsp70-2) and 14-3-3zeta in bladder urothelial carcinoma (BUC) tissues and to explore their roles in tumor development and their clinical significance. Methods Totally 48 specimens of bladder urothelial carcinoma and 20 specimens of bladder urothelial carcinoma were collected. Immunohistochemical SP method, Western blot and reverse transcription-polymerase chain reaction (RT-PCR) were used to detect the expression of Hsp70-2,14 -3-3zeta protein expression at the level of mRNA expression, analysis of the two and bladder urothelial carcinoma pathological grade, clinical stage and other factors. Results All three methods showed that Hsp70-2 and 14-3-3zeta were low expressed in adjacent tissues and highly expressed in bladder urothelial carcinoma tissues. The expression of Hsp70-2 increased with the degree of pathological grading and the depth of invasion. The differences were statistically significant (P <0.05). The expression of Hsp70-2 and 14-3-3zeta protein in bladder urothelial carcinoma were significantly increased (r = 0.516, P <0.05). Conclusions Hsp70-2 and 14-3-3zeta are highly expressed in bladder urothelial carcinoma tissues, which are positively correlated with pathological grade and clinical stage, and also positively correlated with each other, suggesting that they play an important role in tumorigenesis and progression effect.
