目的:探讨水通道蛋白4(aquaporin-4,AQP4)对人结肠腺癌细胞增殖、迁移的影响。方法 :以血管内皮生长因子(VEGF)与AQP4-siRNA分别上调或下调人结肠癌细胞株LoVo细胞AQP4表达,Western blot检测AQP4表达变化;以细胞划痕试验及CCK-8增殖试验分别检测细胞迁移及增殖能力。结果:VEGF可上调LoVo细胞中AQP4的表达,且呈浓度和时间依赖性;VEGF促进LoVo细胞迁移。AQP4-siRNA干扰下调AQP4后细胞迁移能力明显下降;与VEGF处理组比较,AQP4-siRNA与VEGF联合处理后LoVo细胞迁移能力下降。单独应用VEGF或AQP4-siRNA对细胞增殖能力无明显影响。结论:AQP4对人结肠腺癌细胞迁移能力具有促进作用,提示AQP4可能是结肠癌侵润、转移的重要因素之一。
Objective: To investigate the effect of aquaporin-4 (AQP4) on the proliferation and migration of human colon adenocarcinoma cells. Methods: The expression of AQP4 in LoVo cells was up-regulated or down-regulated by VEGF and AQP4-siRNA respectively, and the expression of AQP4 was detected by Western blot. The cell migration was detected by cell scratch assay and CCK-8 proliferation assay And proliferation ability. Results: VEGF could up-regulate the expression of AQP4 in LoVo cells in a concentration-and time-dependent manner. VEGF promoted the migration of LoVo cells. AQP4-siRNA down-regulated AQP4 cell migration significantly decreased compared with the VEGF treatment group, AQP4-siRNA and VEGF combined treatment of LoVo cells decreased migration ability. VEGF or AQP4-siRNA alone had no significant effect on cell proliferation. Conclusion: AQP4 can promote the migration of human colon adenocarcinoma cells, suggesting that AQP4 may be one of the important factors of invasion and metastasis of colon cancer.
