目的探讨姜黄素对原代培养的兔结膜上皮细胞增殖的影响,为预防翼状胬肉术后复发探讨新的策略。方法制备原代兔结膜上皮细胞,以不同浓度的姜黄素(0、10、20、40、60μmol/L)作用于体外培养的第3~5代结膜上皮细胞,分别于处理后24、48、72h后采用MTT法检测细胞增殖情况。此外,采用上述浓度姜黄素处理结膜上皮细胞24h后,采用流式细胞仪检测细胞周期变化。结果 20μmol/L以上浓度的姜黄素可以显著抑制兔结膜上皮细胞增殖,且随浓度增加效果增强,随着时间增加抑制率显著增加(P<0.05)。此外,姜黄素作用24h后的细胞周期变化结果显示,G0/G1期细胞数量增加,细胞增殖受到显著抑制(P<0.05)。结论姜黄素能够抑制体外培养的原代兔结膜上皮增殖,姜黄素有可能作为抑制翼状胬肉术后复发的药物。
Objective To investigate the effect of curcumin on the proliferation of primary cultured rabbit conjunctival epithelial cells and to explore a new strategy for the prevention of postoperative recurrence of pterygium. Methods Primary rabbit conjunctival epithelial cells were prepared and treated with different concentrations of curcumin (0, 10, 20, 40 and 60 μmol / L) for 3 to 5 passages of conjunctival epithelial cells cultured in vitro for 24, 48, 72h after MTT assay cell proliferation. In addition, the above concentration of curcumin conjunctival epithelial cells after 24h, the use of flow cytometry to detect cell cycle changes. Results Curcumin at a concentration of more than 20 μmol / L could significantly inhibit the proliferation of rabbit conjunctival epithelial cells. The inhibitory effect of curcumin increased with increasing concentration (P <0.05). In addition, the results of cell cycle changes after curcumin treatment for 24 h showed that the number of cells in G0 / G1 phase increased and cell proliferation was significantly inhibited (P <0.05). Conclusion Curcumin can inhibit the proliferation of primary rabbit conjunctival epithelial cells cultured in vitro. Curcumin may be used as a drug to inhibit the recurrence of pterygium surgery.
