采田间患病小麦叶片,经PEG沉淀、20%蔗糖垫(含0.3%TritonX-100)离心和氯化铯平衡密度梯度离心获得提纯病毒。紫外扫描呈典型核蛋白吸收曲线,A260:A280为1.15。病毒粒子呈线条状,长度为200~2000nm。病毒核酸为两个组分。免疫电镜观察,小麦梭条斑花叶病毒(WSSMV)抗血清和小麦黄花叶病毒(WYMV)抗血清均能强烈的“修饰”该病毒。提纯病毒免疫家兔,所得病毒抗血清效价可达1/1024,ELISA检测病毒灵敏度达0.128μg·mL-1。
The diseased wheat leaves were harvested from the field and purified by PEG precipitation, centrifugation on a 20% sucrose pad (containing 0.3% Triton X-100) and cesium chloride balance density gradient centrifugation. UV scan showed a typical nuclear protein absorption curve, A260: A280 was 1.15. Viral particles were linear, the length of 200 ~ 2000nm. The viral nucleic acid is two components. Immunoelectron microscopy, wheat splotches mosaic virus (WSSMV) antiserum, and wheat yellow mosaic virus (WYMV) antiserum both strongly “modified” the virus. Purified virus was used to immunize rabbits. The titer of virus antiserum was 1/1024. The sensitivity of ELISA was 0.128μg · mL-1.
