在研究肺表面活性物质层和肺泡Ⅱ型上皮细胞板层体结构的变化过程中,常常由于人工造成的微细结构的丢失而影响实验效果。为了克服这一现象。我们在制片过程中参考了有关资料对如何保存和清晰显示肺泡Ⅱ型上皮细胞板层体的板层结构进行了初步探讨,配制了一种新型的混合固定液,用子肺组织的固定,然后在脱水、包埋之前用醋酸铀水溶液处理。结果表明,用此方法处理的肺组织,不仅肺泡表面活性物质得到良好的显示,而且Ⅱ型上皮细胞内的板层体的板层结构也清晰可见,效果比常规方法有明显提高。材料与方法:将大鼠放血处死,取肺组织按电镜制样要求切成小块,分别浸在二组不同的固定液中。第一组固定液按Luft方法制备,小块组织在“钌红一液”中浸泡1小时,接着用“钌红二液”浸泡3小时,
