目的探讨凝血酶在体内促进肺癌Glc-82移植瘤的发展及其作用机制。方法细胞黏附试验和细胞迁移试验观察凝血酶对Glc-82细胞黏附特性和转移特性的影响,裸鼠移植瘤实验观察凝血酶对Glc-82移植瘤荷瘤小鼠体质量、瘤质量、肺结节及生存期的影响,Western印迹法观察凝血酶对移植瘤组织细胞外信号调节激酶(ERK)、黏着斑激酶(FAK)和抑癌蛋白PTEN(10号染色体上检出的磷酸酶和张力蛋白同源蛋白)的影响。结果凝血酶在体外显著促进Glc-82细胞的黏附和迁移;在体内,凝血酶可降低荷瘤小鼠的体质量、增加瘤质量占体质量比、部分凝血酶组小鼠肺转移增多、荷瘤小鼠的生存时间缩短;Western印迹结果显示,凝血酶可促进肿瘤组织ERK的激活,促进FAK的表达,并且抑制抑癌基因PTEN的表达。结论凝血酶促进肺癌Glc-82移植瘤的恶性进展,可能与促进肿瘤细胞的黏附和迁移特性,促进肿瘤组织ERK、FAK的激活及表达有关,并可能通过抑制抑癌蛋白PTEN的表达而发挥作用。
Objective To investigate the role of thrombin in promoting the development of Glc-82 transplanted lung cancer in vivo. Methods The effects of thrombin on the adhesion and metastasis of Glc-82 cells were observed by cell adhesion assay and cell migration assay. The effects of thrombin on the growth and metastasis of Glc-82 cells in nude mice were observed. The effects of thrombin on the expression of extracellular signal-regulated kinase (ERK), focal adhesion kinase (FAK) and tumor suppressor protein PTEN (phosphatase and tensin protein detected on chromosome 10) were observed by Western blotting. Homologous protein). Results Thrombin significantly promoted the adhesion and migration of Glc-82 cells in vitro. In vivo, thrombin decreased the body weight of tumor-bearing mice, increased the tumor mass to body mass ratio, increased lung metastasis in part of thrombin mice, The survival time of tumor-bearing mice was shortened. Western blotting showed that thrombin promoted the activation of ERK, promoted the expression of FAK and inhibited the expression of tumor suppressor gene PTEN. Conclusion Thrombin can promote the malignant progression of Glc-82 in lung cancer, which may be related to promoting the adhesion and migration of tumor cells, promoting the activation and expression of ERK and FAK, and may play a role in inhibiting the expression of tumor suppressor protein PTEN .
