失血性休克-再灌注大鼠胃黏膜内皮素及一氧化氮含量的变化及丹参提取物F的作用

目的:探讨丹参提取物F(Dan Shen Extract F,DSEF)在胃黏膜缺血-再灌注损伤中的保护作用与胃黏膜内皮素(Endothelin,ET)和一氧化氮(Nitric oxide,NO)的关系。方法:选用Wistar雄性大鼠随机分4组,假手术组(SC组,n=9):进行放血前的所有处理步骤,静脉泵入生理盐水;单纯失血组(HE组,n=9):放血后维持血压在2.67~4.00kPa,20min后处死,静脉泵入生理盐水;生理盐水组(NS组,n=10)和丹参提取物F组(DSEF组,n=12)均回输全部血液,维持20min后处死,NS组静脉泵入生理盐水,DSEF组泵入DSEF。结果:1)各组大鼠失血量及平均动脉压组间无显著差异(P>0.05),而休克前和再灌注后的平均动脉压组内也无显著差异(P>0.05);2)HE组大鼠未见胃黏膜有出血性损伤,DSEF组的损伤指数明显低于NS组(P<0.01);3)NS组NO含量明显低于SC组(P<0.01),DSEF组NO含量明显高于NS组(P<0.01)。NS组ET含量明显增高,与SC组比有明显差异(P<0.01),DSEF组ET含量明显降低,与NS组比有明显差异(P<0.01)。结论:1)胃黏膜NO含量下降,ET含量增加可能是其再灌注损伤的原因。2)DSEF通过维持胃黏膜NO与ET稳定,起到防止胃黏膜-再灌注损伤的作用。 Objective: To investigate the protective effect of Dan Shen Extract F (DSEF) on gastric mucosal ischemia-reperfusion injury and its relationship with endothelin (ET) and nitric oxide (NO) . Methods: Male Wistar rats were randomly divided into 4 groups: sham operation group (SC group, n = 9): all the treatment steps before bloodletting, After bloodletting, blood pressure was maintained at 2.67 ~ 4.00kPa and then was killed after 20min. The rats were infused with normal saline. All the blood was returned to normal saline group (NS group, n = 10) and Salvia miltiorrhiza extract group F (DSEF group, n = 12) , Maintained for 20min after sacrifice, NS group intravenous saline, DSEF group pumped DSEF. Results: 1) There was no significant difference in blood loss and mean arterial pressure between the two groups (P> 0.05), but there was no significant difference between mean arterial pressure before shock and after reperfusion (P> 0.05) There was no gastric mucosal hemorrhagic injury in the HE group, but the damage index in the DSEF group was significantly lower than that in the NS group (P <0.01). 3) The NO level in the NS group was significantly lower than that in the SC group (P <0.01) Significantly higher than NS group (P <0.01). The level of ET in NS group was significantly higher than that in SC group (P <0.01). The ET level in DSEF group was significantly lower than that in NS group (P <0.01). Conclusions: 1) The content of NO in gastric mucosa decreased, and the increase of ET content may be the reason of its reperfusion injury. 2) DSEF plays a role in preventing gastric mucosal-reperfusion injury by maintaining stable gastric mucosal NO and ET.
