目的:探究大孔吸附树脂法分离纯化补阳还五汤中总黄酮的最佳工艺条件和参数。方法:通过静态吸附及解吸试验、动态吸附及洗脱试验,从D101,DA201,AB-8和聚酰胺树脂中筛选出适合补阳还五汤总黄酮分离的理想树脂。并通过单因素分析考察该树脂分离、纯化补阳还五汤总黄酮的最佳工艺条件。结果:经D101大孔吸附树脂纯化后的补阳还五汤总黄酮纯度最高,达到63.1%,转移率为80.2%,静态吸附容量为66.28 mg·g-1(干树脂),最佳动态上样量为20 mL·g-1(总黄酮质量浓度:3.033 g·L-1),上样流速为1 BV·h-1,洗脱乙醇体积分数为60%,洗脱速度为1.5 BV·h-1。结论:D101大孔吸附树脂可较好地用于补阳还五汤总黄酮的分离纯化。
Objective: To explore the optimum conditions and parameters of separation and purification of total flavonoids in Buyang Huanwu Decoction by macroporous adsorption resin method. Methods: The ideal resin suitable for the separation of total flavonoids from Buyang Huanwu Decoction was screened from D101, DA201, AB-8 and polyamide resin by static adsorption and desorption test, dynamic adsorption and elution test. The optimal conditions for the separation and purification of total flavonoids from Buyang Huanwu Decoction were investigated by single factor analysis. Results: The purity of total flavonoids of Bu Yang Huanwu Decoction purified by D101 macroporous resin was the highest, reaching 63.1%, the transfer rate was 80.2%, the static adsorption capacity was 66.28 mg · g-1 (dry resin), the best dynamic The sample volume was 20 mL · g-1 (total flavone mass concentration: 3.033 g · L-1), the sample flow rate was 1 BV · h-1, the volume fraction of ethanol was 60% and the elution rate was 1.5 BV · h-1. Conclusion: D101 macroporous resin can be better used for the separation and purification of total flavonoids in Buyang Huanwu Decoction.
