目的:探讨survivin基因真核表达载体的构建,研究它在新疆哈萨克族食管癌中的作用。方法:采用PCR方法从新疆哈萨克族食管癌的cDNA表达阳性的标本中扩增出survivin基因,经双酶切后、与真核表达载体pEGFP-N1相连后转入DH5α。结果:DNA测序证明获得了包含ORF全长的survivin基因,并成功克隆入真核表达载体pEGFP-N1中。结论:构建真核表达载体成功,为进一步探讨survivin基因在哈萨克族食管癌中的作用奠定了基础。
Objective: To investigate the construction of the eukaryotic expression vector of survivin gene and its role in esophageal cancer of Kazak in Xinjiang. Methods: The survivin gene was amplified by PCR from the positive expression of esophageal cancer in Xinjiang Kazak and cloned into the eukaryotic expression vector pEGFP-N1 and transferred into DH5α. Results: DNA sequencing proved that the full-length survivin gene containing ORF was obtained and successfully cloned into eukaryotic expression vector pEGFP-N1. Conclusion: The successful construction of eukaryotic expression vector lays the foundation for the further study on the role of survivin gene in Kazakh esophageal cancer.
