目的研究苯[α]芘对人B淋巴细胞株2个短串联重复序列位点的遗传效应,探讨人细胞体外系统的遗传稳定性和敏感性。方法采用聚合酶链反应(PCR)分析人B淋巴细胞株2个短串联重复序列位点FGA和D18S379的扩增产物。结果经500、50、5和0.5μmol/L苯[α]芘分别处理细胞株,未发现FGA和D18S379位点发生杂合性丢失,未发现这2个位点出现等位获得,也没有发现重复单位增加。结论在2个目标位点FGA和D18S379没有发生遗传多态性变化。
Objective To investigate the genetic effect of benzene (α) pyrene on two short tandem repeat sites of human B lymphocyte cell line, and to investigate the genetic stability and sensitivity of human cell in vitro. Methods The amplification products of FGA and D18S379 at two short tandem repeat sites of human B lymphocyte were analyzed by polymerase chain reaction (PCR). Results The cell lines were treated with 500, 50, 5 and 0.5 μmol / L Benzo [a] pyrene, respectively. No loss of heterozygosity was found at FGA and D18S379 sites, and no allele was found at these two sites. Repeat unit increase. Conclusion There was no genetic polymorphism in FGA and D18S379 at two target sites.
