目的探讨8号染色体数目异常与FAK蛋白表达在胃肠道间质瘤(GIST)中的关系及其与胃肠道间质瘤预后的关系。方法应用荧光原位杂交(FISH)技术检测石蜡包埋39例GIST中8号染色体的异常情况;免疫组织化学方法检测39例GIST中FAK蛋白的表达情况,结合GIST危险度分级方案进行分析。结果8号染色体发生多倍体者17例,单倍体4例,8号染色体异倍体与GIST复发和转移之间差异有统计学意义(P<0.05);8号染色体异倍体在高级别与低级别GIST间差异有统计学意义(P<0.05)。FAK阳性表达率为49%,与复发和转移之间分别比较差异有统计学意义(P<0.05),高级别GIST中FAK表达高于低级别GIST(P<0.05)。8号染色体多倍体与FAK蛋白阳性表达之间有相关性(r=0.385,P=0.016)。结论GIST中存在8号染色体的数目异常,多倍体多于单倍体,并且与GIST复发转移有关;8号染色体多倍体可能存在靶向基因FAK的位点,8号染色体异倍体与FAK蛋白表达可能是预测GIST复发转移的有用指标。
Objective To investigate the relationship between the number of chromosome 8 abnormality and FAK protein expression in gastrointestinal stromal tumors (GIST) and its relationship with the prognosis of gastrointestinal stromal tumors. Methods Fluorescence in situ hybridization (FISH) was used to detect the abnormality of chromosome 8 in 39 cases of GISTs. The expression of FAK protein in 39 cases of GIST was detected by immunohistochemical staining and analyzed according to the grading scheme of GIST. Results There were 17 polyploidy cases on chromosome 8 and 4 haploids. The difference between aneuploidy on chromosome 8 and recurrence and metastasis of GIST was statistically significant (P <0.05). On the 8th chromosome, The difference between grade and low grade GIST was statistically significant (P <0.05). The positive expression rate of FAK was 49%, which was significantly different from the recurrence and metastasis (P <0.05). The expression of FAK in high grade GIST was higher than that in low grade GIST (P <0.05). There was a correlation between polyploidy on chromosome 8 and FAK protein expression (r = 0.385, P = 0.016). Conclusions The number of chromosome 8 in GIST is abnormal. The number of chromosomes is more than that of haploids, and it is related to the recurrence and metastasis of GIST. The location of FAK on chromosome 8 may exist. The chromosome 8 aneuploidy FAK protein expression may be a useful indicator of GIST recurrence and metastasis.
