目的:从转录因子与细胞因子的角度分析喉鳞状细胞癌患者Th1/Th2细胞分化趋势,探讨T-bet/GATA3与p53的相关性,了解其与肿瘤转移之间的关系。方法:利用荧光定量PCR技术检测49例喉鳞状细胞癌患者(实验组)及30例正常人(正常对照组)外周血单个核细胞(PBMC)中T-bet、GATA3和IFN-γ、IL-4mR-NA的水平;应用免疫组织化学法检测实验组中p53的表达状况。结果:实验组T-bet、GATA3、IFN-γ、IL-4的表达率依次为42.86%(21/49)、71.43%(35/49)、26.53%(13/49)、63.27%(33/49)。定量PCR结果显示,实验组的T-bet和IFN-γ的表达明显低于正常对照组(P<0.05),而GATA3和IL-4的表达则明显高于正常对照组(P<0.05)。在p53阳性的患者中,T-bet的表达量较低,而GATA3的表达量较高,且T-bet和GATA3的表达与p53具有相关性。结论:喉鳞状细胞癌患者有明显的Th2漂移现象,且T-bet与GATA3可作为反映肿瘤转移的参考指标。
OBJECTIVE: To analyze the differentiation trend of Th1 / Th2 cells in laryngeal squamous cell carcinoma from the perspective of transcription factors and cytokines, and to explore the relationship between T-bet / GATA3 and p53 and their relationship with tumor metastasis. Methods: T-bet, GATA3 and IFN-γ in peripheral blood mononuclear cells (PBMCs) of 49 patients with laryngeal squamous cell carcinoma (experimental group) and 30 normal controls (normal control group) were detected by real- -4mR-NA levels; immunohistochemistry was used to detect the expression of p53 in the experimental group. Results: The expression rates of T-bet, GATA3, IFN-γ and IL-4 in the experimental group were 42.86% (21/49), 71.43% (35/49), 26.53% (13/49) and 63.27% / 49). Quantitative PCR results showed that the expression of T-bet and IFN-γin the experimental group was significantly lower than that in the normal control group (P <0.05), while the expression of GATA3 and IL-4 in the experimental group was significantly higher than that in the normal control group (P <0.05). In p53-positive patients, T-bet expression is low, while GATA3 expression is high, and the expression of T-bet and GATA3 is correlated with p53. Conclusions: Th2 shift in laryngeal squamous cell carcinoma is obvious, and T-bet and GATA3 can be used as a reference index to reflect tumor metastasis.
