目的:观察加味藿朴夏苓汤干预对温病湿热证模型大鼠舌组织水通道蛋白-5(AQP-5)的影响。方法:24只SD大鼠,随机分为正常对照组、湿热模型组、药物干预组。参照多因素复合造模方法复制湿热证大鼠模型。光镜下观察舌组织病理改变,采用免疫组化方法检测AQP-5的表达情况。结果:湿热模型大鼠有腻苔形成,药物干预组腻苔出现率较湿热模型组明显减少(P<0.05);湿热模型组大鼠舌黏膜层的角质细胞层、棘细胞层增厚,不完全角化细胞增多;舌固有层组织充血水肿;药物干预组与正常对照组光镜下无明显差别。湿热模型组舌组织AQP-5表达较正常对照组明显增加(P<0.01);药物干预组AQP-5表达较湿热模型组显著减少(P<0.05,P<0.01)。结论:加味藿朴夏苓汤能显著改善温病湿热证模型大鼠腻苔和舌组织病理改变,其机制可能与减轻舌组织炎症,降低AQP-5表达有关。
Objective: To observe the effect of Jiapuxiaolingtang decoction on aquaporin-5 (AQP-5) in tongue tissue of warm-damp heat syndrome rats. Methods: Twenty-four SD rats were randomly divided into normal control group, damp heat model group and drug intervention group. Reference to multi-factor compound modeling method damp-heat rat model. The pathological changes of tongue tissue were observed under light microscope. The expression of AQP-5 was detected by immunohistochemistry. Results: The damp-heat model rats formed greasy moss, and the appearance rate of greasy moss in the drug-treated group was significantly lower than that in the damp-heat model group (P <0.05). In the damp-heat model group, the thickening of keratinocyte layer and spinous cell layer in the tongue mucosa of rats Completely keratinocytes increased; lamina propria congestion hyperemia; drug intervention group and normal control group, no significant difference under light microscope. The expression of AQP-5 in the hot and humid model group was significantly higher than that in the normal control group (P <0.01). The expression of AQP-5 in the model group was significantly decreased compared with the model group (P <0.05, P <0.01). CONCLUSION: Jiapiaoshuling Decoction can significantly improve the pathological changes of greasy moss and tongue in warm damp-heat syndrome rats. The mechanism may be related to the reduction of inflammation and the decrease of AQP-5 expression.
