目的 探讨细胞因子基因多态性与其血清水平的关系。方法 随机选取 65名汉族健康成人 ,分析其肿瘤坏死因子 α(TNF α)、转化生长因子 β1 (TGF β1 )和白细胞介素 1 0 (IL 1 0 )基因型及分泌水平。细胞因子基因型检测采用序列特异引物聚合酶链反应 (PCR SSP)法 ,血清细胞因子水平测定采用ABC ELISA法。结果 TNF α等位基因型别共有 3种 ,分别为A/A、G/A和G/G ,对应的血清TNF α浓度 ,A/A组和G/A组均比G/G组高 (P <0 .0 1 ) ;TGF β1等位基因型别共有 9种 ,分别为T/T G/G、T/C G/G、T/C G/C、C/C G/G、T/T G/C、C/C G/C、C/C C/C、T/T C/C和T/C C/C ,对应的血清TGF β1浓度 ,T/T G/G、T/C G/G组比T/C G/C、C/C G/G和T/T G/C组高 (P <0 .0 1 ) ,而T/C G/C、C/C G/G和T/T G/C组又比C/C G/C、C/C C/C、T/T C/C和T/C C/C组高 (P<0 .0 1 ) ;IL 1 0等位基因型别共有 6种 ,分别为GCC/GCC、GCC/ACC、GCC/ATA、ACC/ACC、ACC/ATA和ATA/ATA ,对应的血清IL 1 0浓度 ,GCC/GCC组比GCC/ACC和GCC/ATA组高 (P <0 .0 1 ) ,而GCC/ACC和GCC/ATA组比ACC/ACC、ACC/ATA和ATA/ATA组高 (P <0 .0 1 )。结论 在受测人群中 ,细胞因子基因分泌型是血清细胞因子水平高低的决定性因素之一
Objective To investigate the relationship between cytokine gene polymorphism and serum levels. Methods Totally 65 Han healthy adults were selected and their genotypes and secretion levels of tumor necrosis factor α (TNFα), transforming growth factor β1 (TGF β1) and interleukin 10 (IL 1 0) were analyzed. Cytokine genotypes were detected by PCR-SSP method and ABC ELISA method was used to detect serum cytokine levels. Results There were three TNFα allele types, which were A / A, G / A and G / G, respectively, corresponding to the serum concentration of TNFα in A / A group and G / A group There were 9 genotypes of TGFβ1 allele, which were T / TG / G, T / CG / G, T / CG / C, C / CG / G, T / TG / C The corresponding serum TGFβ1 concentration, T / TG / G, T / CG / G ratio of T / CG / C C / CG / G and T / TG / C were significantly higher than those in C / CG / C, C / CG / G and T / There were 6 genotypes of IL10 alleles, which were GCC / GCC, GCC / ACC, C / C / C, T / GCC / ATA, ACC / ACC, ACC / ATA and ATA / ATA, corresponding serum IL 1 0 concentrations were higher in GCC / GCC group than in GCC / ACC and GCC / ATA group The ACC and GCC / ATA groups were higher than the ACC / ACC, ACC / ATA and ATA / ATA groups (P <0.01). Conclusions Among the tested population, cytokine secretion is one of the decisive factors in the level of serum cytokines
