目的检测组织蛋白酶-D(Cathepsin D)和细胞核增殖抗原(PCNA)在原发性非小细胞肺癌(NSCLC)中的表达,探讨二者在原发性NSCLC发生、发展中的作用及临床意义。方法采用免疫组化En Vision二步法检测86例原发性NSCLC和25例癌旁正常肺组织中Cathepsin D、PCNA的表达。结果 Cathepsin D在原发性NSCLC组织中的阳性率显著高于正常肺组织(P=0.002),有淋巴结转移组Cathepsin D的表达显著高于无淋巴结转移组(P=0.001);PCNA在原发性NSCLC组织中的阳性率显著高于癌旁正常肺组织(P<0.001),PCNA的高表达与原发性NSCLC的肿瘤临床分期、淋巴结是否转移有关(P=0.031;P=0.033),与组织学分化程度明显有关(P<0.001)。Cathepsin D和PCNA在原发性NSCLC组织中的阳性表达呈正相关(P=0.000,r=0.547)。结论 Cathepsin D和PCNA在NSCLC组织中表达上调,可作为NSCLC研究的重要指标,并对其预后判断有重要意义。
Objective To detect the expression of Cathepsin D and PCNA in patients with primary non-small cell lung cancer (NSCLC) and to explore the role and clinical significance of the two in the development and progression of primary NSCLC. Methods The expression of Cathepsin D and PCNA in 86 NSCLC and 25 normal lung tissues adjacent to normal lung tissues were detected by Envision immunohistochemistry. Results The positive rate of Cathepsin D in primary NSCLC tissues was significantly higher than that in normal lung tissues (P = 0.002). The expression of Cathepsin D in patients with lymph node metastasis was significantly higher than that in patients without lymph node metastasis (P = 0.001) The positive expression rate of PCNA in NSCLC tissues was significantly higher than that in adjacent normal lung tissues (P <0.001). The high expression of PCNA correlated with the tumor stage and lymph node metastasis of primary NSCLC (P = 0.031; P = 0.033) Histological differentiation was significantly related (P <0.001). There was a positive correlation between Cathepsin D and PCNA expression in primary NSCLC (P = 0.000, r = 0.547). Conclusion The up-regulation of Cathepsin D and PCNA in NSCLC tissues may be an important index for the study of NSCLC and is of great significance for the prognosis judgment.
