目的:探讨解毒化瘀中药复方含药血清对白血病K562/A02细胞的耐药逆转作用。方法:应用中药血清药理学方法制备解毒化瘀药的兔血清,以MTT法检测经含药血清、干扰素、川芎嗪处理后K562/A02细胞对化疗药物的敏感性;流式细胞仪检测K562/A02耐药细胞内柔红霉素(DNR)的潴留情况。结果:MTT结果显示,3组药物对K562/A02细胞的耐药逆转倍数分别为9.30倍、5.27倍和3.36倍,其耐药逆转率均在70%以上;流式细胞仪检测结果显示3种药物均可明显增加K562/A02细胞内DNR的浓度,解毒化瘀药含药血清作用最强,与川芎嗪组相比具有显著差异(P<0.01),但与干扰素组相比无明显差异(P>0.05)。结论:3种药物均对K562/A02细胞的耐药具有逆转作用,逆转强度依次为解毒化瘀药含药血清、干扰素、川芎嗪。
Objective: To investigate the reversal effect of decoction-resolving phlegm medicine-containing serum on drug resistance of leukemia K562/A02 cells. Methods: Serum of rabbits detoxified with peony was prepared by serum pharmacology of traditional Chinese medicine. The sensitivity of K562/A02 cells treated with drug-containing serum, interferon, and tetramethylpyrazine to chemotherapeutic drugs was measured by MTT assay; K562 was detected by flow cytometry. Retention of daunorubicin (DNR) in /A02-resistant cells. RESULTS: MTT results showed that the reversal multiples of resistance to K562/A02 cells in the three groups were 9.30-fold, 5.27-fold, and 3.36-fold, respectively, and the drug resistance reversal rate was over 70%; flow cytometry results showed three kinds of results. Drugs can significantly increase the concentration of DNR in K562/A02 cells, and the detoxification and stasis-resolving drug-containing serum has the strongest effect. Compared with the Ligustrazine group, there is a significant difference (P<0.01), but there is no significant difference compared with the interferon group. (P>0.05). Conclusion: All three drugs have a reversal effect on the resistance of K562/A02 cells. The reversal strengths are detoxification and stasis drug-containing serum, interferon, and tetramethylpyrazine.
