目的:观察补肾调冲方对半乳糖致卵巢早衰(Premature ovarian failure,POF)大鼠始基卵泡募集及卵泡闭锁相关因子生长分化因子-9(Growth differentiation factor-9,GDF-9)表达的影响。方法:应用Real-time PCR法检测各组大鼠卵巢内GDF-9基因表达水平;采用Western Blot法检测各组大鼠卵巢内GDF-9蛋白表达水平。应用TUNEL法检测各组大鼠卵巢内细胞凋亡情况。结果:与空白组比较,模型组卵巢内的GDF-9 mRNA与蛋白表达水平明显减少;与模型组比较,治疗组卵巢内的GDF-9基因与蛋白表达明显增加。结论:补肾调冲方可通过调节GDF-9 mRNA与蛋白的表达,促进POF大鼠始基卵泡募集,促进生长卵泡发育,抑制卵泡异常闭锁,从而达到促进排卵,调节卵巢功能,治疗POF的目的。
Objective: To observe the effects of Bushen Tiaochong Recipe on the initiation of primordial follicles and the expression of growth differentiation factor-9 (GDF-9) in follicular atresia-induced injury in rats with premature ovarian failure (POF) induced by galactose. Methods: Real-time PCR method was used to detect the expression of GDF-9 gene in ovary of each group. The expression of GDF-9 protein in ovary of each group was detected by Western Blot. TUNEL method was used to detect the apoptosis of ovary in each group. Results: Compared with the blank group, the expression of GDF-9 mRNA and protein in the ovary of the model group decreased significantly. Compared with the model group, the expression of GDF-9 gene and protein in the ovary increased significantly. Conclusion: Bushen Tiaochong Recipe can promote ovulation, regulate ovarian function and treat POF by regulating the expression of GDF-9 mRNA and protein, promoting primordial follicle recruitment, promoting the growth of growth follicles and inhibiting the abnormal follicular atresia in POF rats.
