为探讨精子表面蛋白受精素 β(Fβ)在精 卵结合和融合中的作用。本文应用鼠Fβ 单抗 ,通过体外受精试验观察该单抗预孵育不同浓度精子后 ,对精 卵结合和融合功能的影响 ;应用免疫细胞化学方法确定Fβ 在精子表面的特异性定位。结果在不同精子浓度下 ,Fβ 单抗均可显著降低体外精 卵的结合和融合能力 ;可使精 卵结合指数降低 34 .43%~ 75 .46 % ,平均 5 6 .0 5 % ;使精 卵融合的受精指数降低 47.6 8%~ 5 6 .32 % ,平均 5 2 .18% ;受精率下降 2 8.0 6 %~5 4.32 % ,平均 41.5 6 %。在精子顶体反应前后 ,Fβ 在精子表面均特异性定位于精子头部赤道区域和顶体内膜区域。结论 :Fβ 在精 卵的结合和融合中均具有重要作用 ,支持将Fβ 蛋白作为精子靶抗原之一用于避孕疫苗的开发研究
To explore the role of sperm surface protein beta (Fβ) in sperm-egg binding and fusion. In this paper, the mouse Fβ monoclonal antibody was used to observe the effect of the monoclonal antibody on sperm-egg binding and fusion after in vitro fertilization test. The immunofluorescence method was used to determine the specific localization of Fβ on sperm surface. Results In different sperm concentrations, Fβ monoclonal antibody significantly reduced the binding and fusion ability of sperm in vitro. The sperm-egg combination index decreased 34.43% -75.46% with an average of 56.05% Fertilization index of fertilized eggs decreased by 47.6 8% ~ 56.32% with an average of 52.18%. The fertilization rate decreased by 28.6% ~ 5.42%, an average of 41.56%. Before and after sperm acrosome reaction, Fβ was located on the sperm surface both in the equatorial region of the sperm head and in the amentoplasty region. CONCLUSION: Fβ plays an important role in sperm-egg binding and fusion, and supports the development of contraceptive vaccines using Fβ as one of the sperm target antigens
