目的 :探讨热休克蛋白 70 (HSP 70 )在癫痫大鼠脑组织内的表达情况及其意义。方法 :采用戊四氮致痫模型 ,应用免疫组织化学及常规病理检查的方法进行研究。结果 :在正常大鼠脑内未见HSP 70免疫反应 (IR)阳性细胞 ,戊四氮致痫后 12h脑内开始出现HSP 70IR阳性细胞 ,2 4hIR达高峰 ,3d后开始下降 ,7d后消失。HSP 70主要在边缘系统 (尤其是海马的CA1、CA3、CA4区 )、大脑皮质 (尤其是颞叶皮质、梨状皮质 )等区域表达。常规病理检查发现 ,上述区域散在出现受损的异常神经元。结论 :癫痫发作可诱导HSP 70在大鼠脑内广泛表达。HSP 70表达可作为神经元受损的一个早期指标。
Objective: To investigate the expression and significance of heat shock protein 70 (HSP 70) in the brain of epileptic rats. Methods: Pentylenetetrazol induced epilepsy model, immunohistochemistry and routine pathological examination methods. RESULTS: No HSP70 immunoreactive (IR) positive cells were found in the normal rat brain. HSP70IR positive cells began to appear within 12 hours after pentylenetetrazol (PTZ) -induced epilepsy, reached the peak at 24 hours and began to decline after 3d and disappeared after 7 days. HSP 70 is mainly expressed in the marginal system (especially in the hippocampus CA1, CA3, CA4 area), cerebral cortex (especially temporal cortex, piriform cortex) and other areas. Routine pathological examination revealed abnormal neurons scattered in the above areas. Conclusion: Epileptic seizures induce the expression of HSP70 in rat brain. HSP 70 expression can be used as an early indicator of neuronal damage.
