目的:研究PTEN和Survivin及HIF-1α蛋白在不同病理分级的胶质瘤中的表达特征及意义。方法:采用免疫组织化学法检测68例脑胶质瘤和10名正常人脑组织标本中PTEN、Survivin和HIF-1α蛋白表达。结果:PTEN蛋白在Ⅰ~Ⅱ级胶质瘤中阳性率为76.47%(26/34),高于Ⅲ~Ⅳ级阳性率为32.35%(11/34),P=0.016;Survivin和HIF-1α蛋白在Ⅰ~Ⅱ级胶质瘤中阳性率为55.88%(19/34)和50.00%(17/34),低于其在Ⅲ~Ⅳ级胶质瘤中阳性表达率为88.24%(30/34)和85.29%(29/34),χ2值分别为8.838和9.676,P均<0.05。PTEN蛋白与Survivin和HIF-1α蛋白的表达均呈负相关,r值分别为-0.504和-0.317,P值分别为0.000和0.008,Survivin蛋白与HIF-1α蛋白的表达呈正相关性,r=0.550,P=0.000。结论:PTEN、Survivin和HIF-1α蛋白表达状态与胶质瘤的分化程度及进展密切相关,联合检测PTEN、Survivin和HIF-1α蛋白有助于判断肿瘤的侵袭性及预后。
OBJECTIVE: To study the expression and significance of PTEN, Survivin and HIF-1α in gliomas of different pathological grades. Methods: The expressions of PTEN, Survivin and HIF-1α in 68 gliomas and 10 normal human brain tissues were detected by immunohistochemistry. Results: The positive rate of PTEN protein in grade Ⅰ ~ Ⅱ glioma was 76.47% (26/34), higher than grade Ⅲ ~ Ⅳ grade was 32.35% (11/34), P = 0.016. Survivin and HIF-1α The positive rates of protein in grade Ⅰ ~ Ⅱ gliomas were 55.88% (19/34) and 50.00% (17/34), which were lower than those in grade Ⅲ ~ Ⅳ gliomas (88.24%, 30 / 34) and 85.29% (29/34) respectively. The χ2 values were 8.838 and 9.676, respectively, P <0.05. The expressions of Survivin and HIF-1α were negatively correlated with the expression of PTEN protein (r = -0.504 and -0.317, P = 0.000 and 0.008, respectively). There was a positive correlation between the expression of Survivin protein and HIF-1α protein , P = 0.000. CONCLUSION: The expression of PTEN, Survivin and HIF-1α is closely related to the differentiation and progression of gliomas. Combined detection of PTEN, Survivin and HIF-1α protein may be helpful in predicting tumor invasiveness and prognosis.
