目的 :研究碱性成纤维细胞生长因子 (bFGF)对大鼠局部脑缺血再灌注后神经功能恢复的影响和机理。方法 :应用线栓法制作Wistar大鼠大脑中动脉闭塞再灌注模型 (MCAO) ,采用行为测试评分观察脑缺血MCAO后的功能恢复情况。结果 :脑缺血MCAO后 1d大鼠患肢感觉运动功能降至最低点 ,随时间延长功能逐渐恢复 ,功能恢复曲线在术后约 5d陡直上升 ,以后缓慢上升 ,至第 2 7d仍未达到正常水平。脑室注射bFGF各时间点行为功能恢复水平高于自然恢复组。前肢试验的恢复高峰在术后第 9d ,前、后肢放置试验到 2 7d与对照组比较差异无统计学意义。后肢试验与平衡木试验在 4周末显示了第 2个恢复高峰。结论 :脑缺血MCAO后肢体功能有自然恢复的趋势 ,外源性bFGF能促使脑缺血MCAO后的肢体功能恢复
Objective: To investigate the effect and mechanism of basic fibroblast growth factor (bFGF) on neural recovery after focal cerebral ischemia / reperfusion in rats. Methods: The middle cerebral artery occlusion and reperfusion model (MCAO) of Wistar rats was made by thread method. The functional recovery after cerebral ischemia was observed by behavioral test score. Results: After 1 week of MCAO, the sensory function of the affected limb decreased to the lowest level and recovered gradually with the prolongation of time. The functional recovery curve rose steeply about 5 days after the operation and then rose slowly until it reached the 27th day normal level. The level of behavioral recovery of bFGF injected into the ventricle at each time point was higher than that of the spontaneous recovery group. The recovery peak of the forelimb test was not significant at the 9th day after the operation, the test of placing the forelimb and hindlimb at 27 days and the control group had no significant difference. The hind limb test and the balance beam test showed the second peak of recovery at 4 weeks. Conclusion: The function of limbs after MCAO has a tendency of natural recovery. Exogenous bFGF can promote the recovery of limb function after MCAO
