圆二色光谱、紫外光谱、红外光谱等光谱技术目前是研究蛋白质、多肽等有机大分子构象与功能关系的有效途径。利用圆二色光谱检测蛋白质并对蛋白的二级结构进行分析、计算,可以得到对蛋白二级结构影响的各种因素和结果。对不同蛋白二级结构的圆二色检测结果表明,在检测过程合适的蛋白浓度对结果的稳定性有非常重要的作用,对于抗体蛋白来说,浓度为10~50μg/mL的区间内,计算得到的二级结构比例是基本稳定的,超出该范围,计算结果偏差加大。研究结果对其他蛋白选择合适浓度进行圆二色检测具有参考意义。
