[目的]分析福建省流行的肠道病毒71(Enterovirus71,EV 71)分离株的遗传特征。[方法]对2008年手足口病例(HFMD)EV 71分离株进行RT-PCR扩增全长VP1基因,测序并分析病毒遗传学特征。[结果]从福州、泉州及龙岩3地市散发的手足口病例中分离到8株EV 71,种系发生树显示2008年福建与阜阳分离株和中国大陆1998年以来的病毒株均为C4亚型,至少分为4个谱系(lineages)。8株福建分离株核苷酸与氨基酸同源性分别为97.4%~100%和99.3%~100%,与Genbank安徽阜阳的5株分离株病毒同源性最高,核苷酸与氨基酸序列同源性为97.6%~99.5%和96.4%~99.3%,其中福建株HF08-17、HS08-140与阜阳分离株同源性为99.2%~99.5%,为同一传播链上的病毒;福清市2分离株(HA08-68和HA08-69)以及长汀县3株(HA08-124,HS08-139和HS08-140)病毒间核苷酸差异性为2.1%~2.2%,说明同一地区存在不同传播链。[结论]2008年福建省EV 71分离株均属于C4亚型,C4亚型病毒在中国大陆具有多传播链、广泛快速传播的特点;福建省内存在亲缘关系较近的EV 71多传播链病毒同时流行,应加强监测与分析。
[Objective] The research aimed to analyze the genetic characteristics of Enterovirus 71 (EV 71) isolated in Fujian Province. [Method] The full-length VP1 gene was amplified by RT-PCR from 2008 HFMD EV71 isolate and sequenced and analyzed for viral genetic features. [Result] Eight EV71 strains were isolated from the cases of hand, foot and mouth emanating from Fuzhou, Quanzhou and Longyan. The phylogenetic trees showed that the isolates of Fuyang and Fuyang in 2008 and the isolates in mainland of China since 1998 were all C4 subunits Type, divided into at least four lines (lineages). The nucleotide and amino acid homology of the isolates in Fujian isolates were 97.4% -100% and 99.3% -100%, respectively, which shared the highest homology with the five isolates from Fuyang, Genbank. The nucleotide and amino acid sequences were homologous The identities of Fujian strains HF08-17 and HS08-140 with Fuyang isolates were 99.2% -99.5%, which were the viruses in the same transmission chain. The isolates of Fuqing city 2 were separated (HA08-68 and HA08-69) and Changting County, the three strains (HA08-124, HS08-139 and HS08-140) were 2.1% -2.2%. This indicated that there were different propagation chains in the same area . [Conclusion] The EV71 isolates belonging to Fujian Province in 2008 belonged to the C4 subtype. The C4 subtype virus had the characteristics of multi-propagation chain in China and spread rapidly and broadly. In Fujian Province, EV 71 multi-transmission chain virus At the same time, monitoring and analysis should be strengthened.
