目的 :了解neuropilin_1基因及其配体semaphorinIII在急性髓性白血病 (AML)骨髓中的表达情况。方法 :利用逆转录_聚合酶链反应扩增20例急性髓性白血病病人骨髓单个核细胞中的neuropilin_1基因及其配体semaphorinIII基因 ,了解其表达情况。14例非血液病患者的骨髓标本作为对照。结果 :急性髓性白血病骨髓单个核细胞中的neuropilin_1基因表达率为15 0% ,semaphorinIII基因表达率为60 0% ,分别与相应正常对照(7 1 % ,57 1% )相比较无显著性差异。结论 :参与调控骨髓基质细胞的neuropilin_1基因及其配体semaphorinIII可表达于AML和正常人的骨髓细胞中 ,该结果为进一步了解此两类基因在造血中的调节作用提供了基础资料
Objective: To investigate the expression of neuropilin_1 gene and its ligand semaphorin III in bone marrow of acute myeloid leukemia (AML). Methods: The gene of neuropilin_1 and its ligand semaphorinIII in bone marrow mononuclear cells from 20 patients with acute myeloid leukemia were amplified by reverse transcription-polymerase chain reaction (RT-PCR) to understand its expression. Bone marrow specimens of 14 non-hematologic patients served as controls. Results: The expression rate of neuropilin_1 gene in bone marrow mononuclear cells of acute myelogenous leukemia was 150% and the expression of semaphorinIII gene was 60%, which was no significant difference compared with the corresponding normal control (71.1%, 57.1%) . CONCLUSION: The neuropilin 1 gene and its ligand semaphorin III, which are involved in the regulation of bone marrow stromal cells, can be expressed in bone marrow cells of AML and normal human. The results provide the basic information for further understanding of the regulation of these two genes in hematopoiesis
