目的:建立用高效液相色谱法同时测定血清中4种苯二氮芯卓类药物氯硝西泮、艾司唑仑、三唑仑、地西泮的方法。方法:血清样品在碱性条件下用乙醚(重蒸)提取,50℃水浴蒸干,残渣用流动相溶解,进样分析。色谱条件为ODS C18柱(5μm,250mm×4.6mm),流动相为甲醇-水-乙腈(53∶42∶5),流速为0.8mL.min-1,检测波长为254nm,柱温为35℃。结果:全部样品洗脱时间均不超过16min,药物之间能完全分离。4种药物平均回收率均在97%以上,样品在各自拟定的质量浓度范围内呈良好的线性关系,最大日内差和日间差分别为4.04%和9.61%。结论:本法提取简单快速,数据可靠,干扰少,适用于苯二氮芯卓类药血药质量浓度检测和中毒患者的急诊诊断。
OBJECTIVE: To establish a method for simultaneous determination of four benzodiazepines, clonazepam, estazolam, triazolam and diazepam in serum by high performance liquid chromatography (HPLC). Methods: Serum samples were extracted with ether (heavy distilled) under alkaline conditions and evaporated to dryness in a water bath at 50 ° C. The residue was dissolved in the mobile phase and analyzed by injection. The chromatographic conditions were ODS C18 column (5μm, 250mm × 4.6mm), the mobile phase was methanol-water-acetonitrile (53:42:5), the flow rate was 0.8mL.min-1, the detection wavelength was 254nm and the column temperature was 35 ℃ . Results: The elution time of all the samples did not exceed 16 min, and the drugs could be completely separated. The average recoveries of the four drugs were above 97%. The samples showed a good linear relationship within the range of their respective mass concentrations. The maximum intraday difference and the intraday difference were 4.04% and 9.61% respectively. Conclusion: The method is simple, rapid, reliable and less interference. It is suitable for the detection of benzodiazepines and the emergency diagnosis of poisoned patients.
