血浆雌三醇放射免疫测定,在临床上对了解胎盘功能、胎儿在宫内发育程度,以及高危妊娠的监护等有一定意义。现将我们建立的测定方法介绍如下: 抗原的制备为了使雌三醇半抗原具有免疫性,须用化学方法制成雌三醇(E_3)与牛血清白蛋白(BSA)的结合物,方可免疫动物使之产生抗体。如要雌三醇抗血清具有高度特异性,就须先将雌三醇转变成雌三醇-6-(O-羧甲基)肟,再和BSA结合,制备出雌三醇-6-牛血清白蛋白抗原,才能用以产生效价高、特异性强的抗血清(图1)。本实验的熔点测定用毛细管法来校正,红外光谱用岛津IR-27G鉴定。
