目的:了解珠海市2007年-2008年H3N2亚型流感病毒流行情况及血凝素基因变异情况。方法:按照时间顺序,随机选取12株2007年-2008年珠海市分离的H3N2亚型流感病毒株进行血凝素HA1序列测定并推导出其氨基酸序列进行分析。结果:H3N2亚型流感病毒在2007年为优势株,2008年成为弱势株。2007年分离株与该年疫苗株比较多数增加了1个糖基化位点,抗原决定簇有4个氨基酸位点发生了替换,并导致其抗原性发生漂移。2008年分离株氨基酸序列与该年疫苗株比较未发生明显变异。结论:2007年根据WHO疫苗推荐株生产的疫苗对该年我市H3N2亚型流感预防效果不好,以致于H3亚型在我市该年度成为优势株,引起广泛流行。2008年H3N2亚型流感病毒为该年度流感流行的弱势株,其HA1基因序列与该年度疫苗株相比未发生明显变异,对人群的保护效果较好。
Objective: To understand the epidemic situation of H3N2 subtype influenza virus and genetic variation of hemagglutinin in Zhuhai from 2007 to 2008. Methods: Twelve hemagglutinin HA1 isolates from 12 isolates of H3N2 isolates isolated in Zhuhai from 2007 to 2008 were randomly selected in chronological order and their amino acid sequences were deduced. Results: H3N2 subtype influenza virus was the predominant strain in 2007 and became the weakest strain in 2008. In 2007, most isolates were increased by one glycosylation site compared with the vaccine strain of the year. There were four amino acid substitutions in the antigenic determinant, leading to the drift of antigenicity. The amino acid sequence of the isolates from 2008 was no significant variation compared with the vaccine strains of the year. Conclusion: The 2007 vaccine produced by the WHO vaccine recommended strains did not have a good effect on the prevention of H3N2 subtype influenza in our city in that year, so that H3 subtype became the predominant strain in our city this year, causing widespread epidemic. H3N2 subtype influenza virus in 2008 was the weakest strain of influenza in this year. The sequence of HA1 gene did not change significantly compared with the vaccine strain in this year, and the protection effect to the population was better.
