从 Mitchell 实验室引进与我们报告的24—26kD 抗原类似的重组 Sj26抗原基因的 E coil,从中抽提并纯化出 rSj26抗原。重组 Sj26抗原初步应用的结果表明:(1)rSj26抗原 ELISA 可以检测日本血吸虫大陆株感染鼠血清中特异性抗体,最高滴度1:320,不比同种24—26kD 抗原检测的效果差;(2)用24—26kD 抗原检测 rsj26免疫鼠血清中的抗体水平,和以 rSj26抗原检测的水平相似,而用 SEA 测抗 rSj26抗体水平较差}(3)用 rSj26免疫鼠后以大陆株尾蚴攻击感染,减虫率可达44.4%,表明 rSj26可诱导一定程度保护性免疫力预防日本血吸虫大陆株尾蚴感染。对 rSj26抗原引进的重要意义进行了讨论。
The E coil of the recombinant Sj26 antigen gene similar to the 24-26kD antigen we reported was introduced from Mitchell Laboratories and the rSj26 antigen was extracted and purified therefrom. The results of preliminary application of recombinant Sj26 antigen showed that: (1) rSj26 antigen ELISA can detect specific antibodies in sera of infected mice with the highest titer of 1: 320, which is no better than that of the same 24-26kD antigen; (2) ) Antibody levels in serum from rsj26-immunized mice were measured using 24-26kD antigen, similar to levels detected with rSj26 antigen, whereas anti-rSj26 antibodies were less detectable with SEA} (3) Inoculation of rSj26 with murine continent cercarid challenge , Worm reduction rate of up to 44.4%, indicating that rSj26 can induce a degree of protective immunity against Schistosoma japonicum cercariae infection. The importance of introduction of rSj26 antigen was discussed.
