目的建立HPLC同时测定凉血通瘀颗粒中芍药苷、丹皮酚、芦荟大黄素、α-细辛醚、大黄素、大黄酚、大黄素甲醚7种成分的方法。方法色谱柱为Lichrospher C18柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱:0~18 min,15%A,体积流量0.8 mL/min;18~20 min,15%~42%A,体积流量0.8 mL/min;20~54 min,42%A,体积流量0.8 mL/min;54~56 min,42%~70%A,体积流量1.0 mL/min;56~80 min,70%A,体积流量1.0 mL/min;检测波长254 nm,柱温25℃。结果 7种成分均能达到基线分离,各成分均有较宽的线性范围和良好的线性关系,加样回收率在95%~105%。结论本法准确、灵敏、可靠、重复性好,可用于凉血通瘀颗粒的质量控制。
OBJECTIVE To establish a HPLC method for simultaneous determination of 7 components of paeoniflorin, paeonol, aloe emodin, α-asarone, emodin, chrysophanol and physcion in Liangxuetongyu Granule. Methods The column was Lichrospher C18 (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile (A) -0.1% phosphoric acid solution (B) with gradient elution: 0-18 min, 15% / min; 18-20 min, 15% -42% A, volumetric flow rate 0.8 mL / min; 20-54 min, 42% A volume flow rate 0.8 mL / min; 54-56 min, 42-70% Volume flow 1.0 mL / min; 56-80 min, 70% A, volume flow 1.0 mL / min; detection wavelength 254 nm, column temperature 25 ℃. Results All the seven components were able to achieve baseline separation. Each component had a wide linear range and a good linear relationship. The recovery rate was between 95% and 105%. Conclusion The method is accurate, sensitive, reliable and reproducible. It can be used for quality control of Liangxuetongyu granules.
