目的:研究板蓝根不同剂型中腺苷的含量,并比较不同制剂间的抗炎作用。方法:采用HPLC测定板蓝根糖浆、板蓝根颗粒、板蓝根软胶囊、板蓝根片、板蓝根滴丸等剂型中腺苷含量,色谱柱为Dia-monsil C18(4.6 mm×200 mm,5μm),检测波长260 nm,流动相甲醇-水(10∶90),流速1.0 mL.min-1,柱温30℃。另将给药后的小鼠,耳廓涂二甲苯致肿胀,比较各组肿胀度差异。结果:腺苷在0.016 0~0.160 0 g线性关系良好(r=0.999 7),平均回收率为98.7%,RSD 0.79%(n=9)。其中板蓝根软胶囊中腺苷含量最高,其他依次为板蓝根颗粒、板蓝根片、板蓝根滴丸、板蓝根糖浆。在二甲苯致小鼠耳廓肿胀的抗炎模型中,板蓝根颗粒抗炎效果最好,其他依次为板蓝根软胶囊、板蓝根滴丸、板蓝根片、板蓝根糖浆。结论:所用方法精密度、重复性良好,结果准确,腺苷含量与药效具有一定的相关性但并不绝对,可能制剂中其他抗炎成分产生了协同作用,应采用多成分指标加以测定或以药效学指标加以控制更为合理。
Objective: To study the content of adenosine in different formulations of Radix Isatidis and to compare the anti-inflammatory effects of different preparations. Methods: The content of adenosine in Radix Isatidis, Radix Isatidis granules, Radix isatidis, Radix Isatidis and Radix Isatidis were measured by HPLC. The detection wavelength was 260 nm with Dia-monsil C18 (4.6 mm × 200 mm, 5 μm) Mobile phase methanol-water (10:90), the flow rate of 1.0 mL.min-1, the column temperature 30 ℃. The other will be administered to mice, auricle coated xylene induced swelling, swelling was compared between groups differences. Results: The linearity of adenosine was 0.016 0 ~ 0.160 0 g (r = 0.999 7). The average recovery was 98.7% and the RSD was 0.79% (n = 9). Including Banlangen soft capsules in the highest content of adenosine, followed by Banlangen particles, Banlangen tablets, Banlangen pills, Banlangen syrup. Among the anti-inflammatory models of xylene-induced mouse ear swelling, Banlangen granules had the best anti-inflammatory effect, followed by Banlangen soft capsules, Banlangen dropping pills, Banlangen tablets and Banlangen syrup. Conclusion: The precision, repeatability and accuracy of the method used are reasonable. The correlation between adenosine content and efficacy is not obvious. It is possible that other anti-inflammatory agents may have a synergistic effect. Multi-component index should be used to determine or Pharmacodynamic indicators to control more reasonable.
