离体冠脉流量实验,是筛选扩冠药物的一个重要方法。近年来,在我国中草药研究工作中,这方面的报道甚多,但在诸多报道中均未注意到灌流液的 pH 值问题,然而灌流液的 pH值对冠流量影响很大。若灌流液 pH 值不合适,离体心脏没有一适宜的化学环境,可导致实验中冠流量不稳定,每分流量锐减太大,因无一稳定基值,而使实验无法进行。偏碱的灌流液能使心肌收缩,小血管床阻力增加,冠流量减少,至 pH 11.0时,心肌处于极度收缩状态,心脏停跳。偏酸的灌流液能使心肌舒张,小血管床阻力下降,冠流量增加,至 pH 2.0时,心肌处于极度舒张状态,心脏停跳。
In vitro coronary flow experiments, is an important method of screening crown drugs. In recent years, there are many reports about Chinese herbal medicine research in this area, but in many reports, no attention has been paid to the pH value of perfusate. However, the pH value of perfusate has a great influence on coronary flow. If the perfusion fluid pH value is inappropriate, the isolated heart does not have a suitable chemical environment, can lead to experimental coronary flow instability, sharp drop in per minute too much, because there is no a stable base value, leaving the experiment can not be carried out. Alkaline perfusion solution can make myocardial contraction, increased resistance to small vascular bed, coronary flow decreased to pH 11.0, the myocardium in an extremely contracted state, cardiac arrest. Partial acid perfusion fluid can make the myocardium diastolic, small vascular bed resistance decreased coronary flow increased to pH 2.0, the myocardium in an extremely relaxed state, cardiac arrest.
