目的:用DNA末端标记法(TUNEL法)研究番荔枝内酯凋亡诱导作用,以及TUNEL法用于凋亡检测和抗癌药所致凋亡的探讨。方法:用台盼蓝拒染法,TUNEL法,DNA凝胶电泳,电镜等技术,研究番荔枝内酯对具有多药抗药性的耐阿霉素的人乳癌MCF7/ADR细胞的生长抑制作用及凋亡诱导。结果:电镜下观察到细胞染色质浓缩,聚集核膜下,核碎裂等典型的凋亡细胞形态学改变。凝胶电泳可见明显的DNA梯形条带。TUNEL法检测的凋亡细胞明显多于形态学改变的凋亡细胞。结论:番荔枝内酯对人乳癌MCF7/ADR细胞具有明显生长抑制作用,半数抑制浓度(IC50)为10.5mg/L,并诱导其凋亡。TUNEL法可配合其他方法用于检测抗癌药诱导的人癌细胞凋亡及抗癌药的评价。
OBJECTIVE: To investigate the apoptosis-inducing effect of annonaceous lactoferrin by DNA terminal labeling (TUNEL) method, and to investigate apoptosis and apoptosis induced by anticancer drugs by TUNEL assay. METHODS: Growth inhibition of human breast cancer MCF-7/ADR cells with a multidrug-resistance of adriamycin was investigated by trypan blue dye exclusion, TUNEL assay, DNA gel electrophoresis and electron microscopy. Effects and induction of apoptosis. RESULTS: Under electron microscopy, the morphology of apoptotic cells such as cell chromatin condensation, subconjunctival nuclear membrane and nuclear fragmentation was observed. Gel electrophoresis shows a distinct DNA ladder. Apoptotic cells detected by TUNEL assay were significantly more than those with morphological changes. Conclusion: Annonaceous lactoferrin has significant growth inhibitory effect on human breast cancer MCF 7/ADR cells with an IC50 of 10.5 mg/L and induces apoptosis. The TUNEL assay can be used in conjunction with other methods to detect anticancer drug-induced apoptosis of human cancer cells and evaluation of anticancer drugs.
