目的探讨牙龈卟啉单胞菌脂多糖(LPS)对人主动脉内皮细胞白细胞介素(IL)-10和单核细胞趋化蛋白(MCP)-1表达的影响。方法用不同浓度(0、10、100、150 ng/ml)的牙龈卟啉单胞菌脂多糖分别作用于人主动脉内皮细胞。酶联免疫吸附法、聚合酶链反应检测IL-10和MCP-1的表达。结果在一定浓度范围内,牙龈卟啉单胞菌LPS呈剂量依赖性促进人主动脉内皮细胞(HAECs)MCP-1的表达,同时减低IL-10的表达水平。结论牙龈卟啉单胞菌LPS可诱导人主动脉内皮细胞IL-10和MCP-1表达紊乱,从而促进动脉粥样硬化的发生。
Objective To investigate the effect of LPS on the expression of interleukin (IL) -10 and monocyte chemotactic protein (MCP) -1 in human aortic endothelial cells. Methods Different concentrations (0, 10, 100, 150 ng / ml) of Porphyromonas gingivalis lipopolysaccharide were administered to human aortic endothelial cells. The expression of IL-10 and MCP-1 were detected by enzyme linked immunosorbent assay and polymerase chain reaction. Results In a certain concentration range, LPS of P. gingivalis promoted the expression of MCP-1 in HAECs in a dose-dependent manner and decreased the expression of IL-10. Conclusion LPS can induce the disorder of IL-10 and MCP-1 expression in human aortic endothelial cells and promote the development of atherosclerosis.
