目的:优化合成环孢素A不完全抗原(CsA-BBa)的工艺。方法:采用正交设计法,以环孢素A不完全抗原(CsA-BBa)的得率为测定指标,优化环孢素A(CsA)与4-苯甲酰苯甲酸(BBa)在光化学反应的条件下合成环孢素A不完全抗原的工艺。结果:合成环孢素A不完全抗原(CsA-BBa)的最佳工艺是:CsA与BBa的摩尔比为1∶1.1,光照距离为10 cm,反应溶剂为叔丁醇,反应时间为3 h。结论:优选的最佳条件稳定可行,且工艺方法简便易行,可用于下一步环孢素A完全抗原的制备。
Objective: To optimize the synthetic cyclosporin A antigen (CsA-BBa) process. Methods: The orthogonal design method was used to optimize the determination of cyclosporine A (CsA) and 4-benzoylbenzoic acid (BBa) in the photochemical reaction Under the conditions of cyclosporin A incomplete antigen synthesis process. Results: The optimum conditions for the synthesis of CsA-BBa were as follows: the molar ratio of CsA to BBa was 1: 1.1, the light distance was 10 cm, the reaction solvent was tert-butanol, and the reaction time was 3 h . Conclusion: The optimum conditions are stable and feasible, and the method is simple and easy to use, which can be used for the next step of preparation of CsA complete antigen.
