自1964年以来,我们用尿液水解试验与热盐水溶解试验协助诊断急性白血病的细胞类型,结果满意。方法简单,一般基层医院均可做此试验,故介绍如下: 一、材料与方法 1.尿液水解试验: ①取正常人新鲜尿液,用0.1N盐酸和0.1N氢氧化钠调节PH值至中性或弱硷性,然后加温至60℃±2℃。②将新鲜骨髓片或血片,用甲醇固定。或用瑞氏染色后的涂片,待空气中干燥后,置于上述尿液中15分钟,后移出并用自来水冲洗待干。③再用瑞氏染色法染色(时间宜长15到20分钟),于油镜下观察。 2.热盐水溶解试验: ①取骨髓及血片用甲醇固定,或用瑞氏染色后的涂片。②将固定后的涂片或瑞氏染色后之涂片,放入60℃的生理盐水中10分钟。
We have been able to diagnose the type of acute leukemia cell lines with urine hydrolysis test and hot salt water dissolution test since 1964 with satisfactory results. The method is simple, the general primary hospital can do this test, it is introduced as follows: First, materials and methods 1. Urine hydrolysis test: ① normal fresh urine, 0.1N hydrochloric acid and 0.1N sodium hydroxide to adjust the PH value to Neutral or weakly alkaline, and then warmed to 60 ℃ ± 2 ℃. ② fresh bone marrow slices or blood films, fixed with methanol. Or Watson stained smear, to be air-dried, placed in the urine for 15 minutes, then removed and rinsed with tap water to be dried. ③ Wright staining and then staining (time should be 15 to 20 minutes), observed under an oil microscope. 2. Hot salt water dissolution test: ① bone marrow and blood films were fixed with methanol, or Wright stain smear. ② After the fixed smear or Wright stain smear, into 60 ℃ saline for 10 minutes.
