试验以苍耳子中黄酮为提取目标,优化苍耳子中黄酮的提取工艺。采用索氏提取法提取苍耳子中黄酮类化合物。以甲醇为溶剂,通过单因素试验考察提取时间、料液比和提取温度对黄酮类物质提取率的影响。最优提取工艺为提取溶剂甲醇、提取时间2.0 h、料液比1∶30及温度70℃,在此条件下提取率为2.34%。然后用大孔树脂分离纯化黄酮,最后采用牛津杯法,分别以大肠杆菌和枯草芽孢杆菌为供试菌,研究黄酮对2种菌的抑菌活性。试验结果显示,大肠杆菌的抑菌效果优于枯草芽孢杆菌。
In this study, the extraction of flavonoids from Xanthium sibiricum was carried out to optimize the extraction of flavonoids from Xanthium ssp. Extraction of flavonoids from Xanthium sibiricum with Soxhlet extraction. Using methanol as solvent, the effects of extraction time, solid-liquid ratio and extraction temperature on the extraction rate of flavonoids were investigated by single factor experiments. The optimal extraction process was extraction solvent methanol, extraction time 2.0 h, solid-liquid ratio 1:30 and temperature 70 ℃. Under these conditions, the extraction rate was 2.34%. Then, the macroporous resin was used to separate and purify the flavonoids. Finally, the Oxford cup method was used to study the antibacterial activities of flavonoids against two kinds of bacteria, respectively, using Escherichia coli and Bacillus subtilis as test strains. The test results show that the bacteriostatic effect of Escherichia coli is better than Bacillus subtilis.
