目的筛查血浆微小RNA在乙肝疫苗接种无应答儿童血浆中的差异表达,为寻找预测疫苗应答效应的临床指标提供线索。方法通过采用巢式病例对照方法从参加儿童保健的儿童中确定研究对象,最终获得10名乙肝疫苗接种无应答儿童及10名应答儿童,通过微小RNA芯片技术检测应答组与无应答组中微小RNAs的表达差异,并采用RTPCR验证芯片检测结果。结果与应答组比较,在乙肝疫苗免疫前,无应答组有3个异常表达的微小RNA,在乙肝疫苗再免疫后,无应答组共有5个异常表达的微小RNA。这些异常表达的微小RNA多具有免疫调控功能。结论乙肝疫苗接种无应答儿童存在异常表达的微小RNA,这些微小RNA可能通过对免疫系统的调控而影响乙肝疫苗的接种效应。
Objective To screen the differential expression of plasma microRNAs in the plasma of unresponsive children with hepatitis B vaccination and provide clues for the search of clinical indicators for predicting the vaccine response. Methods The nested case-control method was used to identify the subjects from children who participated in child health care. Ten non-responders and 10 respondents were enrolled in this study. MicroRNAs were used to detect microRNAs The difference between the expression and the RTPCR verification chip test results. Results Compared with the response group, there were three abnormally expressed microRNAs in the non-responder group before HBV vaccine immunization. There were five microRNAs abnormally expressed in the non-responder group after re-immunization of hepatitis B vaccine. Many of these abnormally expressed microRNAs have immunomodulatory functions. Conclusion Hepatitis B vaccines inoculated non-responders have abnormally expressed microRNAs. These microRNAs may affect the vaccination effect of hepatitis B vaccine through regulating the immune system.
