研究了采用高效液相色谱 紫外检测法对盐酸赖氨酸注射液中赖氨酸含量的测定 .作者在AminoQuant方法的基础上 ,改为仅采用邻苯二醛 (OPA)衍生化 ,省略了原方法衍生化试剂中的不稳定成分 ,而采用优化的反应条件提高紫外测定的灵敏度 ;又对这个组成较简单的样品改用恒定组成的流动相淋洗 ,手动方法测定避免了对该试样不必要的梯度淋洗 ,较原AminoQuant氨基酸分析法简便、快速 ,结果峰面积与浓度相关系数r =0 9998,回收率为 99 7% (RSD为 1 0 %~ 1 5 % ) .在采用紫外检测法时 ,最小检出量为 5 3× 10 -8(S/N =3)与原方法相近
