目的:研究淫羊藿甙(ICA)对PG细胞分泌转化生长因子β2(transforming growth factorβ2,TGFβ2)的影响、ICA逆转TGFβ2介导的免疫抑制作用及其机制。方法:MTF法检测细胞增殖、杀伤活性,RT-PCR检测TGFβ2、穿孔素,流式细胞术检测TGFβ2、IL-2Rα水平。结果:ICA可抑制PG细胞中TGFβ2的蛋白和mRNA表达,能够逆转受TGFβ2抑制的LAK和CD3AK细胞的杀伤活性,并能部分恢复受TGFβ2抑制的LAK细胞表面IL-2Rα表达和CD3AK细胞内穿孔素mRNA水平,以及CD3AK细胞的增殖活性。结论:淫羊藿甙通过抑制肿瘤细胞免疫抑制因子TGFβ2的产生,增强免疫效应细胞的杀伤活性等机制而发挥抗肿瘤作用。
Objective: To study the effect of epimedium (ICA) on the secretion of transforming growth factor β2 (TGFβ2) in PG cells and the mechanism of ICA reversing the immunosuppression induced by TGFβ2. Methods: The cell proliferation and killing activity were detected by MTT assay. TGFβ2 and perforin were detected by RT-PCR. The levels of TGFβ2 and IL-2Rα were measured by flow cytometry. RESULTS: ICA inhibited the expression of TGFβ2 protein and mRNA in PG cells, reversed the cytotoxic activity of TGFβ2 inhibiting LAK and CD3AK cells, and partially restored the expression of IL-2Rα on the surface of LAK cells inhibited by TGFβ2 and perforin in CD3AK cells. mRNA levels, and proliferation activity of CD3AK cells. Conclusion: Epimedium exerts anti-tumor effect by inhibiting the production of tumor cell immunosuppressive factor TGFβ2 and enhancing the killing activity of immune effector cells.
