本实验在Wistar糖尿病大鼠模型上,研究微囊化新生猪胰岛样细胞团(ICC)的功能存活,以及移植后纠正糖尿病大鼠高血糖状态的能力.新生猪胰腺经Ⅺ型胶原酶消化后培养,每只猪可获得2500~3000个10c.以海藻酸钠-多聚赖氨酸-海藻酸钠膜包裹后再培养,ICC在囊内继续分化成熟.微囊化ICC分泌胰岛素在20天内维持一定水平,而单纯ICC在2周后基本不能测出胰岛素.糖刺激实验,微囊化ICC较单纯ICC无明显差异.体内实验中,微囊化ICC移植组8只大鼠,每只大鼠移植2000~3000个囊(含ICC5000个左
In this study, we investigated the functional survival of microencapsulated neonatal porcine islet-like cell mass (ICC) and its ability to correct hyperglycemia in diabetic rats after Wistar diabetic rat model.After the piglets were digested with Ⅺ-type collagenase Culture, each pig can be obtained 2500 ~ 3000 10c.With sodium alginate - polylysine - sodium alginate membrane wrapped and then cultured, ICC continue to differentiate and mature in the cyst.Individual insulin secretion in ICC within 20 days While ICC could not detect insulin after 2 weeks.It was found that there was no significant difference of ICC between ICC and ICC in sugar stimulated experiment.In in vivo experiment, 8 rats in ICC transplantation group, each big Mouse transplantation 2000 ~ 3000 capsules (including ICC5000 left
