目的探讨ezrin和CD44-v6在食管鳞癌中的表达及其与淋巴结转移,病理学分级等的关系。方法应用组织芯片技术进行免疫组化S-P法染色,检测71例食管鳞癌患者癌组织标本及癌旁正常上皮中ezrin和CC44-v6的表达情况,并结合临床相关因素进行统计学分析。结果71例癌旁正常上皮中,ezrin表达阳性33例,CD44-v15表达阳性18例;71例食管鳞癌组织中,ezrin表达阳性64例,CD44-v6表达阳性58例。Ezrin的表达与淋巴结转移关系密切,有淋巴结转移组ezrin强阳性率明显高于无淋巴结转移组(P<0.05);CD44-v6的表达与肿瘤组织的病理学分级、淋巴结转移关系密切,在淋巴结转移组CD44-v6表达阳性率显著高于无淋巴结转移组(P<0.05)。CD44-v6阳性表达组的ezrin表达阳性率显著高于CD44-v6阴性表达组(P<0.05)。Ezrin与CD44-v6同时表达的48例患者中,淋巴结转移率高达60.0%,而ezrin与CD44-v6同时不表达的6例患者中,均无淋巴结转移。结论联合检测ezrin和CD44-v6的表达有助于综合判断食管鳞癌的恶性程度和转移潜能,ezrin有望成为判断肿瘤预后的新指标。
Objective To investigate the expression of ezrin and CD44-v6 in esophageal squamous cell carcinoma and its relationship with lymph node metastasis and pathological grade. Methods Tissue microarray was used to perform immunohistochemical S-P staining. The expression of ezrin and CC44-v6 in 71 specimens of esophageal squamous cell carcinoma and adjacent normal epithelium were detected. The results were statistically analyzed by combining clinical factors. RESULTS: In 71 cases of normal epithelium, ezrin was positive in 33 cases, and CD44-v15 was positive in 18 cases. In 71 cases of esophageal squamous cell carcinoma, ezrin was positive in 64 cases, and CD44-v6 was positive in 58 cases. The expression of Ezrin was closely related to lymph node metastasis. The strong positive rate of ezrin in lymph node metastasis group was significantly higher than that in no lymph node metastasis group (P<0.05). The expression of CD44-v6 was closely related to the pathological grade and lymph node metastasis of tumor tissues, and was closely related to lymph nodes. The positive rate of CD44-v6 expression in the metastatic group was significantly higher than that in the no lymph node metastasis group (P<0.05). The positive rate of ezrin expression in CD44-v6 positive expression group was significantly higher than that in CD44-v6 negative expression group (P<0.05). Among the 48 patients whose Ezrin and CD44-v6 were simultaneously expressed, the lymph node metastasis rate was as high as 60.0%, while no lymph node metastasis was found in 6 patients who did not express both ezrin and CD44-v6. Conclusion The combined detection of ezrin and CD44-v6 expressions is helpful for the comprehensive evaluation of the malignant degree and metastatic potential of esophageal squamous cell carcinoma. ezrin is expected to become a new index for judging the prognosis of cancer.
