目的比较恬尔心和硝苯地平对慢性铁过负荷大鼠肝、肾和脾组织铁沉积与营养代谢的影响及探究可能的作用机制。方法雄性SD大鼠通过喂饲高铁饲料,制备慢性铁过负荷动物模型。动物腹腔分别注射一定剂量的恬尔心和硝苯地平后,组织学染色观察肝、肾和脾组织铁沉积;同时测定血清中营养代谢相关指标的含量。结果恬尔心组与模型组比较,肝、肾和脾组织中铁黄素沉积未明显降低,甘油三脂(TG)由(1.86±0.75)mmol/L降至(1.54±0.45)mmol/L,高密度脂蛋白(HDL)由(0.67±0.21)mmol/L降至(0.5±0.25)mmol/L。硝苯地平组与模型组比较,肝、肾和脾组织中铁黄素沉积明显降低,总胆固醇(TC)由(1.91±0.34)mmol/L降至(1.65±0.36)mmol/L,低密度脂蛋白(LDL)由(1.14±0.15)mmol/L降低到(0.86±0.16)mmol/L,丙二醛(MDA)由(15.27±3.61)μmol/L降至(11.86±3.52)μmol/L,血清中铁(Fe)含量由(1.57±0.25)mg/L降至(1.32±0.14)mg/L,血清中硒(Se)含量由(0.072±0.012)mg/L升至(0.087±0.017)mg/L,钙(Ca)含量由(76.87±15.20)mg/L升至(97.25±16.47)mg/L。结论恬尔心对铁过负荷大鼠营养代谢的治疗效果不明显,低于硝苯地平的疗效。
Objective To compare the effects of tian er heart and nifedipine on the iron deposition and nutrient metabolism in the liver, kidney and spleen of chronic iron overload rats and explore the possible mechanism. Methods Male SD rats were fed with high-iron diet to prepare chronic iron overload animal model. The animals were injected intraperitoneally with a certain dose of tian er heart and nifedipine respectively, histological staining was used to observe the iron deposition in the liver, kidney and spleen tissues. At the same time, the content of serum nutrition metabolites was measured. Results Compared with model group, the deposition of lutein in liver, kidney and spleen was not significantly reduced in TG group and the triglyceride (TG) decreased from (1.86 ± 0.75) mmol / L to (1.54 ± 0.45) mmol / L, High density lipoprotein (HDL) decreased from (0.67 ± 0.21) mmol / L to (0.5 ± 0.25) mmol / L. In nifedipine group, the deposition of lutein in liver, kidney and spleen decreased significantly from (1.91 ± 0.34) mmol / L to (1.65 ± 0.36) mmol / L in low dose group compared with model group (LDL) decreased from (1.14 ± 0.15) mmol / L to (0.86 ± 0.16) mmol / L and MDA decreased from (15.27 ± 3.61) μmol / L to (11.86 ± 3.52) μmol / The content of Fe in serum decreased from (1.57 ± 0.25) mg / L to (1.32 ± 0.14) mg / L and the content of selenium in serum increased from (0.072 ± 0.012) mg / L to (0.087 ± 0.017) mg / L, and the content of calcium increased from (76.87 ± 15.20) mg / L to (97.25 ± 16.47) mg / L. Conclusion TITEL heart treatment of iron overload rats nutrition metabolism effect is not obvious, lower than the efficacy of nifedipine.
