目的:阐明肺纤维化过程中肺成纤维细胞(LF)产生透明质酸(HA)的动态变化及作用。方法:检测和对比博莱霉素(BLM)致大鼠肺纤维化不同时期支气管肺泡灌洗液(BALF)和LF培养上清中HA水平。结果:(1)BLM组LF培养上清中HA水平于第1天明显升高,第3天达高峰,第14~28天时降至正常。(2)BLM组BALF中HA水平于第3天明显升高,第7天达高峰,随后下降,但至第14天时仍高于正常水平,第28天降至正常。(3)BLM组BALF和LF上清中HA水平与BALF细胞成分之间均具有明显的正相关;BALF和LF培养上清中HA水平之间也有明显的相关性。结论:LF在肺纤维化的早期肺泡炎阶段就处于活化状态,产生HA增多,导致肺内HA的过多聚积;LF产生HA增多和BALF中HA水平的增高可反映肺泡炎的强度和疾病的活动性
Objective: To clarify the dynamic changes of hyaluronic acid (HA) produced by pulmonary fibroblasts (LF) during the process of pulmonary fibrosis. Methods: The level of HA in bronchoalveolar lavage fluid (BALF) and LF culture supernatant of bleomycin (BLM) -induced pulmonary fibrosis in rats at different stages were detected and compared. Results: (1) In the BLM group, the level of HA in LF culture supernatant increased significantly on the first day, peaked on the third day, and returned to normal on the 14th to 28th day. (2) The level of HA in BALF of BLM group increased significantly on the 3rd day and peaked on the 7th day, then decreased, but still higher than the normal level on the 14th day and dropped to normal on the 28th day. (3) There was a significant positive correlation between HA level in BALF and LF supernatant of BLM group and cellular components of BALF. There was also a significant correlation between HA level in BALF and LF culture supernatant. CONCLUSIONS: LF is activated during the early phase of alveolitis of pulmonary fibrosis, resulting in increased HA, resulting in excessive accumulation of HA in the lung. Increased HA in LF and increased HA in BALF may reflect the intensity and severity of alveolitis Activity
