[Objective] To determine the content of chlorogenic acid in extracts of FLOS LONICERAE JAPONICAE at different flowering periods that were prepared by different method.[Method] The chlorogenic acid(CA) in FLOS LONICERAE JAPONICAE was determined and identified by UV spectrophotometric method after extraction with ethanol reflux method or ultrasonic method,aiming at discussing the difference of chlorogenic acid at different flowering periods.[Result] The concentration and content of chlorogenic acid in original extract of FLOS LONICERAE JAPONICAE prepared by ethanol reflux method were 1.905 mg/ml and 3.81%,while those in original extract prepared by supersonic method were 1.306 mg/ml and 2.61%.Compared with supersonic method,ethanol reflux method was optimized and validated.Moreover,the concentration and content of chlorogenic acid in original extract of L.japonica full-flowering flowers prepared by ethanol reflux were 0.167 7 mg/ml and 0.34%,while whose in original extract of L.japonica buds were 0.260 1 mg/ml and 0.52%.Hence,the bud contained more chlorogenic acid.[Conclusion] The method is easy,simple and rapid to extract chlorogenic acid and determine its content in FLOS LONICERAE JAPONICAE.
