The most frequent gram-negative bacteria that are resistant to tetracyclines are triggered by the drug recognition of Tet repressors. It causes a repressor-operator DNA complex association that enables the expression of the drug resistant protein TetA, which is responsible for the active efflux of tetracycline. The 2.5 A resolution crystal structure of Tet repressor tetracycline-magnesium complex dimer reveals detailed drug recognition. The direction of the repressor’s manipulation and the associated helix-corner-helix structure are reversed compared to other DNA-binding proteins. Due to its DNA-binding structure than pass
