目的:探讨靶向人端粒酶基因hTERT对人喉鳞状细胞癌Hep-2细胞生长增殖的抑制作用。方法:根据人端粒酶基因hTERT设计和合成人端粒酶siRNA和随机的siN.C作为阴性对照,通过转染载体Lipofectamine 2000脂质体转染Hep-2细胞,进行形态学、RT-PCR和MTT法检测,观察其干扰效果。结果:①siRNA能够成功转染进入Hep-2细胞;②RT-PCR结果显示转染组与阴性对照组和空白组相比,hTERT mRNA表达明显减弱,转染组hTERT mRNA较空白组相对下降70%;③形态学和MTT结果显示人端粒酶siRNA能有效抑制Hep-2细胞增殖,抑制率达到65%。结论:靶向hTERT的siRNA能显著抑制喉鳞状细胞癌Hep-2细胞的生长增殖。
Objective: To investigate the inhibitory effect of targeting human telomerase gene hTERT on the growth and proliferation of human laryngeal squamous cell carcinoma Hep-2 cells. Methods: The human telomerase gene hTERT was designed and synthesized. Human telomerase siRNA and random siN.C were used as negative control. Hep-2 cells were transfected with lipofectamine 2000 liposome and the morphology and RT-PCR And MTT test to observe the interference effect. Results: ① siRNA was successfully transfected into Hep-2 cells; ② The results of RT-PCR showed that the expression of hTERT mRNA in transfection group was significantly decreased compared with the blank control group and the blank control group. The hTERT mRNA in transfection group was decreased by 70% ③ Morphology and MTT results showed that human telomerase siRNA can effectively inhibit the proliferation of Hep-2 cells with the inhibition rate of 65%. Conclusion: siRNA targeting hTERT can significantly inhibit the growth and proliferation of laryngeal squamous cell carcinoma Hep-2 cells.
