目的 :制备抗重组人肝再生增强因子 (rhALR)单克隆抗体 (McAb)。方法 :以纯化的rhALR为抗原免疫小鼠 ,取其脾细胞与SP2 /0细胞融合 ,以ELISA ,Westernblot法分别筛选测其滴度及免疫特异性。结果 :获得 2株稳定分泌抗rhALR单克隆抗体杂交瘤株。其效价 ,腹水 10 -3 - 10 -5,培养上清 10 -2 - 10 -3 ,两株McAb针对不同抗原决定簇。结论 :制备的抗体具有高特异性。对rhALR在体内组织器官的分布 ,示踪rhALR在体内的代谢及探索rhALR在翻译水平上对细胞发育的调控分布等研究具有重要的应用价值
Objective: To prepare anti-recombinant human monoclonal antibody (rhALR) for human liver regeneration (McAb). Methods: The purified rhALR was used as antigen to immunize mice. The spleen cells were fused with SP2 / 0 cells. The titers and immunospecificities of SP2 / 0 cells were determined by ELISA and Western blot respectively. Results: Two strains of monoclonal antibodies against rhALR were successfully secreted. The titer was 10 -3 -10 -5 in ascites and 10 -2 -10 -3 in culture supernatant. The two McAbs were directed against different antigenic determinants. Conclusion: The prepared antibodies have high specificity. The distribution of rhALR in the tissues and organs of the body, tracing the metabolism of rhALR in vivo and exploring the regulatory distribution of rhALR on the cell development at the translation level are of great value
