目的:筛选黄芪、三七组成的通脉复方有效组分,并确定其最佳配伍剂量。方法:SD大鼠随机分为空白血清组、金水宝组1.32 g·kg-1、通脉水煎8 g·kg-1组、通脉提取物8 g·kg-1组、复方多糖0.64 g·kg-1组、复方皂苷0.16 g·kg-1组、其余组分组,共7组。ig给药7 d,7 d后采集大鼠含药血清。采用四甲基偶氮唑盐(MTT)比色法测定不同提取组分含药血清对肾小球系膜细胞(GMC)增殖的影响。在此基础上,采用2因素3水平的正交试验L9(34)对“有效组分”多糖、皂苷最佳配伍组合进行了研究。结果:正常培养及LPS刺激条件下,复方多糖与复方皂苷、通脉提取物、水煎液、金水宝含药血清均能抑制GMC增殖,与空白对照组相比,均有显著或非常显著性差异(P<0.05~0.01);而其余组分含药血清对GMC增殖无明显抑制,与空白对照组相比无显著性差异;通脉提取物含药血清对GMC增殖的抑制作用强于复方多糖、复方皂苷及金水宝含药血清,组间比较,有显著或非常显著性差异(P<0.05~P<0.01)。对主效应即GMC增殖的抑制作用的贡献程度(F值)复方多糖(13.69)与复方皂苷(13.67)几乎无差别。有效组分不同剂量配伍中,以A3B3即复方多糖960 mg、复方皂苷240 mg配伍对GMC的抑制作用最为显著。结论:复方多糖与复方皂苷为通脉复方的主要有效组分,其比例为4:1(960 mg:240 mg)为有效组分最佳剂量配伍。
Objective: To screen the effective components of Tongmai which composed of Astragalus and Panax notoginseng, and determine the best compatible dose. Methods: SD rats were randomly divided into blank serum group, Jinbao group 1.32 g · kg -1, Tongmai decoction 8 g · kg -1 group, Tongmai extract 8 g · kg -1 group, compound polysaccharide 0.64 g · Kg-1 group, compound saponin 0.16 g · kg-1 group, the rest of the group, a total of 7 groups. ig administration 7 d, 7 d after the collection of rat serum. MTT assay was used to determine the effect of different extract fractions on the proliferation of glomerular mesangial cells (GMCs). On this basis, the optimum combination of “effective components”, “polysaccharides and saponins” was studied by orthogonal experiment L9 (34) with 2 factors and 3 levels. Results: Under the conditions of normal culture and LPS stimulation, compound polysaccharides and compound saponins, Tongmai extract, Decoction and Jinshuibao serum could inhibit the proliferation of GMC, both significantly and very significantly compared with the blank control group (P <0.05-0.01). However, the drug-containing serum of the other components had no significant inhibition on the proliferation of GMC, which showed no significant difference compared with the blank control group. The serum containing Tongmai extract inhibited the proliferation of GMC significantly Polysaccharides, compound saponins and Jinshuibao containing serum, there was significant or very significant difference between groups (P <0.05 ~ P <0.01). The degree of contribution (F value) to the main effect, which is the inhibitory effect of GMC proliferation, was almost indistinguishable from compound saponin (13.67). The effective components of different doses compatibility, with A3B3 compound polysaccharide 960 mg, compound saponin 240 mg compatibility of GMC inhibition the most significant. Conclusion: The compound polysaccharides and compound saponins are the main effective components of Tongmai compound, the ratio of which is 4: 1 (960 mg: 240 mg), and the best dosage is compatibility.
